A universal 6iL/E4 culture system for deriving and maintaining embryonic stem cells across mammalian species
Abstract The derivation of authentic embryonic stem cells (ESCs) across mammalian species remains a major challenge. Here, we report the development of a defined, serum-free culture system, termed 6iL/E4, that enables the derivation and long-term self-renewal of ESCs across diverse mammalian species. Through systematic dissection of signaling pathways, we identified conserved regulatory modules involving GSK3α, WNT, STAT3, PDGFR, and MEK/ERK signaling.

Dear Editor, Pluripotent stem cells (PSCs) generate derivatives of all three germ layers; however, robust contribution to chimeric embryos is largely restricted to early pluripotent states.

Tripeptide DT-109 (Gly-Gly-Leu) attenuates atherosclerosis and vascular calcification in nonhuman primates - Signal Transduction and Targeted Therapy
Abstract Advanced atherosclerotic lesions and vascular calcification substantially increase the risk of cardiovascular events. However, effective strategies for preventing or treating advanced atherosclerosis and calcification are currently lacking. This study investigated the efficacy of DT-109 (Gly-Gly-Leu) in attenuating atherosclerosis and calcification in nonhuman primates, exploring its broader therapeutic potential.
Vascular smooth muscle cell PRDM16 regulates circadian variation in blood pressure
Abstract Disruptions of blood pressure (BP) circadian variation are closely associated with an increased risk of cardiovascular disease. Thus, gaining insights into the molecular mechanisms of BP circadian variation is essential for comprehending BP regulation. Human genetic analyses suggest that PR domain–containing protein 16 (PRDM16), a transcription factor highly expressed in vascular smooth muscle cells (VSMCs), is significantly associated with BP-related traits.
Blood pressure regulation through circadian variation: PRDM16 as a target in vascular smooth muscle cells
Abstract The precise mechanisms of blood pressure (BP) regulation are not fully elucidated, and understanding BP regulation is crucial for managing hypertension and improving outcomes for cardiovascular disease. In this issue of the JCI, Wang et al. identified the transcription factor PR domain–containing protein 16 (PRDM16) as a regulator of both vascular smooth muscle cell contraction and the circadian response to BP control.
Annexin A1 binds PDZ and LIM domain 7 to inhibit adipogenesis and prevent obesity - Signal Transduction and Targeted Therapy
Abstract Obesity is a global issue that warrants the identification of more effective therapeutic targets and a better understanding of the pivotal molecular pathogenesis. Annexin A1 (ANXA1) is known to inhibit phospholipase A2, exhibiting anti-inflammatory activity. However, the specific effects of ANXA1 in obesity and the underlying mechanisms of action remain unclear. Our study reveals that ANXA1 levels are elevated in the adipose tissue of individuals with obesity.

Recent progress in polymeric gene vectors: Delivery mechanisms, molecular designs, and applications
A. Mechanisms of gene delivery by polymeric gene vectors The brief mechanism of polymer mediated gene delivery is illustrated in Fig. 1, including the formation of the polyplexes (gene packaging), entering cells via endocytosis, endosome/lysosome escape, gene release, entering the nucleus (for plasmid DNA), and expression.
Gut microbiota production of trimethyl-5-aminovaleric acid reduces fatty acid oxidation and accelerates cardiac hypertrophy - Nature Communications
Abstract Numerous studies found intestinal microbiota alterations which are thought to affect the development of various diseases through the production of gut-derived metabolites. However, the specific metabolites and their pathophysiological contribution to cardiac hypertrophy or heart failure progression still remain unclear.
