Extended follow-up of in vivo BCMA CAR-T therapy in relapsed/refractory multiple myeloma
Abstract Data on long-term outcomes of in vivo chimeric antigen receptor T cell (CAR‑T) therapy remain scarce. We previously reported preliminary safety and efficacy data of in vivo B-cell maturation antigen (BCMA) CAR‑T (ESO-T01), in four patients with relapsed/refractory multiple myeloma, without leukapheresis and lymphodepletion, with up to 3 months of follow-up.
Adsorption sequence shapes antigen distribution and antibody binding on adjuvants in bivalent vaccines
Abstract As a critical advancement in modern immunization strategies, multivalent vaccines combine multiple antigenic components to seamlessly broaden protection against diverse pathogens within a single dose. During the formulation of aluminum-adjuvanted vaccines, antigen adsorption onto the adjuvant surface serves as a critical determinant of vaccine performance.

Advances in the Biosynthesis of Noncanonical Amino Acids for Genetic Code Expansion
1 Introduction The discoveries of selenocysteine [1] and pyrrolysine [2-5] fundamentally reshaped our understanding of the genetic code, which was long believed to encode only 20 amino acids in nearly all organisms. In methanogenic archaea, pyrrolysine, the 22nd proteinogenic amino acid, is biosynthesized directly from lysine [6] and incorporated into proteins through a dedicated translational machinery analogous to that used for canonical amino acids (cAAs).
Digital inclusion, legal governance, and the adoption of smart care services by older adults in China: an evolutionary game analysis
Abstract Introduction: The availability of digital care services does not ensure their adoption by older adults. Methods: This study develops a tripartite evolutionary game involving government, platform enterprises, and older adults to examine how legal governance, enterprise investment, and user-side benefits jointly shape the adoption of smart care services by older adults in China.

Transplantation of human pluripotent stem cell-derived pericytes ameliorates radiation-induced brain injury via CCR5 signaling
Abstract Radiation-induced brain injury remains an intractable complication of cranial radiotherapy, characterized by disrupted blood-brain barrier, inflammation, and progressive neuronal loss. Pericytes sustain severe damage in this pathology, yet no reliable repair treatment exists. Here we show that human pluripotent stem cell-derived pericytes exert therapeutic benefits in a mouse model of gamma knife radiation brain damage.

Invisible harm and constrained response: a conceptual analysis of psychological domestic abuse against disabled people in China
Abstract Psychological domestic abuse against disabled people is under-recognised at the intersection of violence prevention, public mental health, and disability policy. Here, invisible harm does not mean wholly unseen harm; it denotes cumulative abuse that remains difficult for victims, social networks, and institutions to identify or translate into action. Constrained response denotes the linked restrictions on recognition and disclosure, evidentiary translation, and safe exit.
T-cell engager bispecific antibodies enable T cells to eliminate Multiple Myeloma by targeting tumor CD38 in combination with PD-L1
Abstract Bispecific antibodies have gradually been applied in the treatment of multiple myeloma(MM). The combination of bispecific antibodies targeting CD3 and CD28 mimics the dual-signaling system (antigen signaling and co-stimulatory signaling) of natural T cell activation, thereby significantly enhancing the anti-tumor response of T cells. CD38, a transmembrane glycoprotein, is highly expressed on MM cells and serves as an important surface marker.
METTL14-dependent N 6 -methyladenosine modification of FGF16 participates in IGFBP5 deficiency-enhanced angiogenesis
Abstract N6-methyladenosine (m6A) RNA modification plays critical roles in physiological and pathological processes. Our prior study demonstrated that IGFBP5 expression is upregulated in the ischemic limb, whereas endothelial-specific IGFBP5 knockout (Igfbp5EKO) protects against hind limb ischemia by enhancing angiogenesis.
How we made GitHub Copilot CLI more selective about delegation
Better orchestration, fewer handoffs, faster progress, without a single new knob. June 12, 2026 | 8 minutes Share: In agentic systems, more delegation isn’t always better. Imagine asking Copilot CLI to make a simple change. Instead of handling it directly, it spins up a helper agent that searches the repository, waits on a result, and stalls. Work that should have taken one step now takes three.
Receptiveness of physicians towards artificial intelligence-driven drug prescription: a nationwide survey
|
Abstract Using artificial intelligence (AI) to prescribe drugs has advanced slowly. Whether a “doctor-in-the-loop” design would increase acceptance of drug-prescribing AI is unknown, as are settings where physicians envision AI-driven drug prescription most likely to be implemented. We surveyed a stratified sample of 2708 physicians throughout China to interrogate their opinions on drug-prescribing AI.

AAV8-mediated mouse/human PROC expression rescues thrombophilia in hereditary protein C-deficient mice
|
Abstract Hereditary protein C (PC) deficiency, which is caused by PROC gene mutations, increases the risk of venous thromboembolism and offers limited treatment options. In this study, we developed adeno-associated virus serotype 8 (AAV8) vectors carrying either murine PC (AAV8-mPROC) or human PC (AAV8-hPROC) transgenes. These vectors were delivered through tail vein injection into PROC knockout mice.

Ulcerative colitis-driven gut dysbiosis exacerbates periodontal bone loss through the gut–oral axis /Th17/Treg imbalance
Abstract Ulcerative colitis (UC) and periodontitis, both microbial dysbiosis-driven chronic inflammatory disorders, coexist and mutually exacerbate, but the causal mechanisms remain unclear. Using ligature-induced periodontitis plus DSS-colitis mice, we found UC doubles alveolar bone loss, heightens systemic inflammation, oxidative stress, and osteoclastogenesis.

SHANK3 mutations disrupt olfactory valence coding across species, with cortical amygdala mechanisms identified in mice
Abstract Mutations in SHANK3 are a leading monogenic cause of autism spectrum disorder (ASD), often associated with profound sensory abnormalities. However, the impact of SHANK3 deficiency on olfactory processing and the underlying neural mechanisms remains unclear. Here, we identify a cross-species disruption of olfactory valence perception in individuals with SHANK3 mutations and in Shank3 mutant mice.

Ubiquitination of DDX21 by HERC2 induces a dormancy-like phenotype via the NUCKS1-p21/p27 axis to promote radio-resistance in colorectal cancer cells
Abstract Cellular dormancy in colorectal cancer (CRC) significantly contributes to therapeutic resistance, tumor recurrence, and metastasis, resulting in poor prognosis. However, the underlying molecular mechanisms remain poorly understood. Here, we used spheroid culture combined with serum deprivation to enrich and identify dormancy-like CRC cells in vitro and characterized their dormancy-like phenotype by G0/G1 phase arrest, suppressed proliferation and radio-resistance.

Sequence Display enables large-scale sequence–activity datasets for rapid protein evolution
Abstract Engineering proteins with desired functions remains challenging and usually requires multiple rounds of screening and selection. Here, we present Sequence Display, a platform that generates large-scale protein sequence–activity datasets in a single round. Sequence Display enables multiplexed assessment of individual variant activity within a single experiment, offering a robust approach to mapping detailed sequence–function relationships.
Isotopic Signature of Organic Nitrogen in Authigenic Carbonates: Implications for Dynamic Nitrogen Cycling at Submarine Cold Seeps
1 Introduction The seepage of hydrocarbon-rich and primarily methane-rich fluids from marine sediments into the overlying water column, with the sites known as cold seeps, is widespread along continental margins (Joye, 2020; Suess, 2014). Cold seeps sustain complex ecosystems supported by the anaerobic oxidation of methane (AOM), a process primarily mediated by consortia of anaerobic methanotrophic archaea (ANME) and sulfate-reducing bacteria (Boetius et al., 2000).

The relationship between college students’ legal cognition and maladaptive risk-taking behaviors: the moderating effect of need for cognitive closure
1 Introduction Adolescence is characterized by heightened engagement in maladaptive risk-taking behaviors, such as substance use, reckless driving, and delinquency, which existing frameworks only partially explain (Willoughby et al., 2021; Leather, 2009). Such behaviors are associated with adverse developmental outcomes, including academic underachievement, mental health problems, and future antisocial tendencies (McLeod et al., 2012).

Real-time imaging of protein microenvironment changes in cells with rotor-based fluorescent amino acids - Nature Chemical Biology
Abstract Fluorescent protein fusions with environmentally sensitive fluorophores have been widely used to investigate changes in the protein microenvironment. Unfortunately, these techniques often rely on bulky fluorescent proteins or tags to the N terminus or C terminus of the target protein, which can disrupt the behavior of the target protein and may limit their ability to investigate microenvironment changes with high spatial resolution.

Innovative rapid screening for complex BCR-ABL1 kinase domain mutations in TKI-treated leukaemia patients - British Journal of Cancer
Abstract Screening for BCR-ABL1 kinase domain (KD) mutations is routinely implemented in cases of treatment failure for chronic myeloid leukaemia and Philadelphia-positive acute lymphoblastic leukaemia. However, timely deciphering their clonal relationship via mutation profiling that requires identification of mutation types, quantification of mutant abundance, and differentiation between compound and polyclonal mutations (CMs and PMs), remains difficult during therapy.
CARD11 signaling regulates CD8+ T cell tumoricidal function - Nature Immunology
Abstract Chronic stimulation in the tumor microenvironment can induce exhausted CD8+ T (Tex) cells that have limited tumoricidal activity. Here, we show an inverse correlation between signal strength and Tex cell differentiation by using patient-derived mutations in the T cell receptor (TCR)-signaling protein CARD11. Strong TCR signaling of the E134G mutant inhibits Tex cell differentiation and increases tumor growth.
