Inflammatory neuropathy in mouse and primate models of colorectal cancer
Abstract Colorectal cancer survivors are at increased risk of developing neurological issues, particularly peripheral neuropathy and chronic pain. Although pre-existing neuropathy is a risk factor for chronic pain, tumor-induced neuropathy has not been firmly established in pre-clinical models. Consistent with clinical observations, we show that mice with colorectal cancer develop peripheral neuropathy, which was associated with subtle locomotor deficits, without overt hypersensitivity.

Abstract Rapid and comprehensive analysis of complex proteomes across large sample sets is vital for unlocking the potential of systems biology. We present a high-throughput mass spectrometry (MS) proteomics method that integrates narrow-window data-independent acquisition (nDIA) with short-gradient micro-flow chromatography, enabling profiling of >240 samples per day. This optimized MS approach identifies 6,201 and 7,466 human proteins with 1- and 2-min gradients, respectively.
Elevated emissions of oviposition‐induced mango volatiles mediate foraging behavior of a parasitoid wasp, Fopius arisanus
1 INTRODUCTION As a highly diverse group of insects, parasitoids play a critical role in the biological control of devastating pests through antagonistic interactions.1 They utilize long-range detectable plant volatile organic compounds (VOCs) as olfactory cues for host location.2 Consequently, parasitoids have evolved an incredible ability to discriminate among different blends of VOCs induced in plants by their herbivore hosts.
Supercharging-enhanced nDIA-MS enables global profiling of drug-induced proteome solubility shifts - Nature Communications
Abstract Mass spectrometry (MS) is indispensable for high-throughput quantitation of protein expression. But protein function is regulated by factors beyond abundance alone. Here, we evaluate two supercharging reagents, dimethyl sulfoxide (DMSO) and m-nitrobenzyl alcohol (mNBA), in narrow-window data-independent acquisition (nDIA)-MS. DMSO markedly enhances MS signal and protein identification, whereas mNBA primarily increases peptide identifications.

Robust Twin Extreme Learning Machine Based on Soft Truncated Capped L1-Norm Loss Function
1. Introduction Extreme learning machine (ELM) [1,2] was proposed by Huang et al. on a single hidden layer feedforward network. Compared with traditional training methods, ELM has advantages in learning speed and generalization performance. This is because ELM differs from traditional methods, that is, the input consists of randomly generated weights and bias values of the hidden layer nodes, allowing for the quick and effective determination of output weights.

Inhibition of hepatic oxalate overproduction ameliorates metabolic dysfunction-associated steatohepatitis - Nature Metabolism
|
Abstract The incidence of metabolic dysfunction-associated steatohepatitis (MASH) is on the rise, and with limited pharmacological therapy available, identification of new metabolic targets is urgently needed. Oxalate is a terminal metabolite produced from glyoxylate by hepatic lactate dehydrogenase (LDHA). The liver-specific alanine-glyoxylate aminotransferase (AGXT) detoxifies glyoxylate, preventing oxalate accumulation.

Minimum loss optimization of flywheel energy storage systems via distributed adaptive dynamic programming
In this article, a distributed controller based on adaptive dynamic programming is proposed to solve the minimum loss problem of flywheel energy storage systems (FESS). We first formulate a performance function aiming to reduce total losses of FESS in power distribution applications. Then we use the Hamilton–Jacobi–Bellman (HJB) equation to solve this optimal control problem. The solution of the HJB equation is approximated by neural networks.
Caspase-1 activates gasdermin A in non-mammals
eLife assessment This study presents valuable insights into the evolution of the gasdermin family, making a strong case that a GSDMA-like gasdermin activated by caspase-1 cleavage was already present in early land vertebrates. Convincing biochemical evidence is provided that extant avian, reptilian, and amphibian GSDMA proteins can still be activated by caspase-1 and upon cleavage induce pyroptosis-like cell death -- at least that they do so in the context of human cell lines.
NLRP3, NLRP6, and NLRP12 are inflammasomes with distinct expression patterns
Abstract Inflammasomes are sensors that detect cytosolic microbial molecules or cellular damage, and in response they initiate a form of lytic regulated cell death called pyroptosis. Inflammasomes signal via homotypic protein-protein interactions where CARD or PYD domains are crucial for recruiting downstream partners. Here, we screened these domains from NLR family proteins, and found that the PYD domain of NLRP6 and NLRP12 could activate caspase-1 to induce cleavage of IL-1β and GSDMD.
