Cancer-induced nerve injury promotes resistance to anti-PD-1 therapy - Nature

Abstract Perineural invasion (PNI) is a well-established factor of poor prognosis in multiple cancer types1, yet its mechanism remains unclear. Here we provide clinical and mechanistic insights into the role of PNI and cancer-induced nerve injury (CINI) in resistance to anti-PD-1 therapy. Our study demonstrates that PNI and CINI of tumour-associated nerves are associated with poor response to anti-PD-1 therapy among patients with cutaneous squamous cell carcinoma, melanoma and gastric cancer.

đźŹ-️ From Nothing to a Wall of Protection: What It Really Means to Be a Founder
By Patrick Dougherty | Founder, Cyber Integrity Solutions Most people think founding a company starts with an idea. But for some of us, it starts with pain. It starts with watching people get hurt because the systems that were supposed to protect them — digital or physical — failed. It starts with being told, “You’re not the right fit,” because you didn’t go to the right school or have the right connections. It starts with seeing security treated like a luxury, not a necessity.
Human pain neuroscience and the next generation of pain therapeutics

Pain Experience and Sensory Changes in Astronauts During and After Short-Lasting Commercial Spaceflight: A Proof-of-Concept Study
Abstract Space travel has been associated with musculoskeletal pain, yet little is known about the nociceptive changes and pain experience during spaceflight. This preliminary study aims to investigate the pain experience and sensory alterations in astronauts following a 17-day mission to the International Space Station (ISS) on Axiom Space's AX-1 commercial space flight. Two participants were enrolled, and data were collected pre-flight, in-flight, post-flight, and three-month post-flight.
The Endosomal Escape Vehicle Platform Enhances Delivery of Oligonucleotides in Preclinical Models of Neuromuscular Disorders
Abstract Biological therapeutic agents are highly targeted and potent but limited in their ability to reach intracellular targets. These limitations often necessitate high therapeutic doses and can be associated with less-than-optimal therapeutic activity. One promising solution for therapeutic agent delivery is use of cell-penetrating peptides.

Building AI Agents: Lessons Learned over the past Year
1. Reasoning is more important than knowledge This quote has echoed in my head over the last 12 months: Agents are the appropriate response to this! And I would apply this logic when building an agent as: Focus less on what your agent “knows”, and more on its ability to “think”. For example, let’s consider writing SQL Queries. SQL queries fail… a lot. In my time as a data scientist, I’m sure I had a lot more queries fail than I ever did succeed.
OPINION: Let’s give competence a chance
Anchorage residents have been put through the wringer the past couple of years. First the trauma and stress of COVID-19, then a cratering economy, chronic homelessness, a seemingly ever-shabbier community and one municipal management crisis after another. But Anchorage is about to catch a break. We are lucky to have a mayoral race where the right choice is obvious, one in which we have an outstanding alternative to a sad status quo.

The young woman, also known as as Jane “Newport” was estimated to be in her mid-teens to early 20’s, and investigators determined that she likely died of suspicious or “foul play” circumstances. Lab results indicated a high probability she died in the late 1960s.
Mel Blanc and the Unsung History of Great Voice Acting
Since the creation of animated motion pictures, the use of actors supplying voices has been invaluable. Behind every famous cartoon character's personality is the actor standing in the recording booth and stretching their manner of speaking in unrecognizable ways, all for the sake of entertainment.
Lightyear and the Curse of Franchise Fatigue
Very few things in the film-going experience match the excitement of knowing that a new entry in your favorite franchise is coming down the pipeline. Big box office sequels to majorly successful movies can more often than not print money when released at just the right time, satiating audiences' appetite for more. However, what happens when even the die-hard fans' attentions begin to wain, and the theater seats stop being sold out?
Explaining the Waning Appeal of Crossovers
The idea of characters from separate shows interacting with one another is a concept dating back decades in the world of television. From classic sitcoms to modern animated cartoons, crossovers remain one of the most common ways for networks to churn up excitement for their programming, with either combining two powerhouse shows into one for major ratings, or using an already popular vehicle to promote another that may be struggling.
The Looney Tunes Show: Awkward Misstep or Clever Reinvention?
With a franchise as big as the Looney Tunes, one can expect there to be countless variations on the characters and formula throughout the decades. While some fair better than others, there have been few interpretations more controversial to fans than 2011's The Looney Tunes Show. Utilizing a more contemporary format for the classic toons, many viewers related the series to that of Seinfeld in its dry humor and focus on a more domesticated atmosphere.
Bubba Ho-Tep 20 Years Later: Remembering When Elvis and JFK Fought a Mummy
There are two types of people in this world: Those who would be interested in watching a film about an elderly Elvis Presley and Black John F. Kennedy defeating an ancient Egyptian mummy, and those who would be incorrect. Luckily for the former, we have the film Bubba Ho-Tep to scratch that eerily specific itch. The 2002 horror comedy film would almost seem as if it were a fever dream when explained to those who have never heard of it.

Home Movies: One of the Funniest Shows Ever Made About Kids
Though an oft-forgotten relic of Adult Swim's colorful past, Home Movies has maintained a die-hard cult following for over two decades since its release. Running for four seasons from 1999 to 2003, the cartoon would revolve around a young boy named Brendan Small and his two friends, Melissa Robbins and Jason Penopolis. The three children would spend their time acting and writing in home movies of their own creation, while also juggling the seemingly impossible task of growing up and coming-of-age.

The Best Animated Series That Got Canceled Too Soon
It's always a tragic thing to see a project made with much love and care be canceled before it can really take off. While there are some series lasting so long that people debate on when it could possibly end (The Simpsons), others meet the unfortunate fate of being snuffed out prematurely. Whether it be from network disputes, poor ratings, bad time slots, or all of the above, fans of animation seem to always be able to cite their own particular examples of a show that ended too soon.
Back to the Future and the Age of Sci-Fi Simplicity
Of all film genres, science fiction has perhaps seen the most growth over the history of the medium. Often heavy in visual effects, the progression of computer generated animation has done wonders for elevating the magnitude and spectacular nature of these fantastical works of fiction. Though the modern age of sci-fi can be home to some of the most inventive imagery and stories, the bygone-yet-beloved era of the genre still has its deep roots of influence, albeit from a simpler time.

Bullethead: Don Rickle's Hilariously Venomous Career
As perhaps one of the final vestiges of the "Borscht Belt" style of comedy that ruled over celebrity roasts and stand-up throughout the previous century, Don Rickles remains without equal, even years after his passing. No one was louder, faster, or more adept at picking apart an unsuspecting victim's slip-ups than the late insult comedian, with talk show appearances and stints on The Dean Martin Celebrity Roast being the prime examples.

Hollywood’s First Scandal: The Tainted Legacy of Fatty Arbuckle
In the current age of seemingly endless gossip and scandals surrounding celebrities, the "gotcha" style of news reporting feels as if it must be a relatively recent advancement in the world of entertainment media. While the level of coverage may be vastly more prominent these days, actors from way back in the silent era of films could still fall prey to the rantings of publication outlets, and the prying eyes of the public who hope to get a front seat to a fascinating fall from grace.
