The psychiatric risk gene CACNA1C encodes distinct full-length isoforms in human brain, heart and aorta
Abstract CACNA1C is a risk gene for multiple psychiatric disorders and the CaV1.2 protein it encodes is a potential target for their treatment. It is known that alternative start exon usage and splicing of CACNA1C RNA results in multiple transcript isoforms. However, the identity of full-length isoforms, and their relative abundance between tissues, remains unknown.
Secondary prevention lipid management after ACS at a DGH: a quality improvement project
Lipid management is a key component of secondary prevention after acute coronary syndrome (ACS), but guideline adherence is variable. Adjunctive lipid-lowering therapies (LLTs) beyond statins are available, yet eligibility in the real-world setting is not well-described.
Spatiomolecular mapping reveals anatomical organization of heterogeneous cell types in the human nucleus accumbens

Results We generated paired spatially resolved (SRT) and single-nucleus (snRNA-seq) transcriptomics data from adjacent tissue sections of 10 adult neurotypical brain donors (6 male, 4 female) (Figure 1A; Table S1; STAR Methods). Anatomical alignment revealed inter-donor variability in sample position along the anterior-posterior (A-P) axis, which facilitated examination of molecular composition across anterior, intermediate, and posterior positions (Figure S1; Data S1).

BDNF-DT and BDNF-AS-DT : novel genes in the BDNF locus
Abstract Divergent transcription from bidirectional promoters is frequently observed in eukaryotic genomes, but the biological relevance of divergent RNA transcripts (DT) is unknown. We identified and characterized BDNF-DT, a novel DT gene, and BDNF-AS-DT, a novel readthrough gene, in the locus containing BDNF, a gene with key roles in neuronal development, differentiation, and synaptic plasticity.

203. Genetic Architecture of MicroRNA Expression in Human Brain Tissues and Its Contribution to Psychiatric Disorders
Large-scale genomic studies demonstrate that genetic variants influence molecular phenotypes, yet the genetic regulation of microRNAs (miRNAs), key post-transcriptional regulators, remain largely underexplored in human brain tissues. Prior miRNA genetic studies are limited by small sample sizes, restricted brain regions, and predominantly European cohorts.
408. Single-Nucleus Atlas of Gene Expression in the Human Brain in African-Ancestry Individuals and Implications for Brain Disorders
Single-cell transcriptomics enables discovery of cell type-specific genetic mechanisms underlying gene regulation and disease vulnerability in the human brain. African-ancestry individuals remain underrepresented in these studies, limiting insight into ancestry-related regulatory diversity relevant to brain disorders.
SETD1A regulates psychiatric gene networks involved in genomic stability and synaptic function in rare and sporadic schizophrenia - Nature Communications
Abstract Rare loss-of-function (LoF) mutations in SETD1A are associated with schizophrenia (SCZ). However, how SETD1A haploinsufficiency leads to SCZ-associated phenotypes and its relevance to patients without these rare mutations is unknown. Here, we identify SETD1A bound loci and regulated genes in human prenatal cortex and isogenic pluripotent stem cell-derived neuronal models engineered with SETD1A LoF variants, including the most common patient mutation.
Budget 2025: Fair for thee but not for me
By Thomas Hyde, First Year, Politics and International Relations The feeling that most dominated this year’s budget was relief: relief for a chancellor who has secured her own job; relief for children being lifted out of poverty; and relief for working people who may well think it could have been worse. Encountering the chancellor at her post-budget celebration, she appeared genuinely pleased with the reaction from MPs, financial markets, and wider society.
Cross-species analysis of adult hippocampal neurogenesis reveals human-specific gene expression but convergent biological processes - Nature Neuroscience
Abstract Immature dentate granule cells (imGCs) arising from adult hippocampal neurogenesis contribute to plasticity, learning and memory, but their evolutionary changes across species and specialized features in humans remain poorly understood. Here we performed machine-learning-augmented analysis of published single-nucleus RNA-sequencing datasets and identified macaque imGCs with transcriptome-wide immature neuronal characteristics.

An integrated single-nucleus and spatial transcriptomics atlas reveals the molecular landscape of the human hippocampus - Nature Neuroscience
Abstract Cell types in the hippocampus with unique morphology, physiology and connectivity serve specialized functions associated with cognition and mood. These cell types are spatially organized, necessitating molecular profiling strategies that retain cytoarchitectural organization. Here we generated spatially-resolved transcriptomics (SRT) and single-nucleus RNA-sequencing (snRNA-seq) data from anterior human hippocampus in ten adult neurotypical donors.

Analysis of gene expression in the postmortem brain of neurotypical Black Americans reveals contributions of genetic ancestry - Nature Neuroscience
Abstract Ancestral differences in genomic variation affect the regulation of gene expression; however, most gene expression studies have been limited to European ancestry samples or adjusted to identify ancestry-independent associations. Here, we instead examined the impact of genetic ancestry on gene expression and DNA methylation in the postmortem brain tissue of admixed Black American neurotypical individuals to identify ancestry-dependent and ancestry-independent contributions.
Dopamine signaling enriched striatal gene set predicts striatal dopamine synthesis and physiological activity in vivo - Nature Communications
Abstract The polygenic architecture of schizophrenia implicates several molecular pathways involved in synaptic function. However, it is unclear how polygenic risk funnels through these pathways to translate into syndromic illness. Using tensor decomposition, we analyze gene co-expression in the caudate nucleus, hippocampus, and dorsolateral prefrontal cortex of post-mortem brain samples from 358 individuals.

Guidelines for Buying Walking Shoes
There are three essential factors that should be taken into consideration before purchasing a new pair of walking shoes. Stability - the shoes should have a balanced and secure feel throughout range of motion. Flexibility - the shoes should allow for a good degree of give at the base of the toes, providing smooth motion. Comfort - walking shoes should comprise contours and padding conformed closely to the feet, providing a snug fit at the heel and midfoot, with ample room in the forefoot.
Transcriptome-scale spatial gene expression in the human dorsolateral prefrontal cortex - Nature Neuroscience
Abstract We used the 10x Genomics Visium platform to define the spatial topography of gene expression in the six-layered human dorsolateral prefrontal cortex. We identified extensive layer-enriched expression signatures and refined associations to previous laminar markers. We overlaid our laminar expression signatures on large-scale single nucleus RNA-sequencing data, enhancing spatial annotation of expression-driven clusters.

How I turned the Profumo scandal into a one-singer opera
I remember the moment I told George Vass that my opera would require only one singer. A large smile appeared on his face and his pen scribbled something down on the proposed budget. My decision to use one voice to play all the ‘characters’ was motivated by a desire to find the most convincing theatrical presentation for my protagonist, Dr Stephen Ward, the osteopath at the centre of the Profumo Scandal in the 1960s.
Facing our fears
Are you anxious? Are you afraid of something? I feel a lot of anxiety around us. We’re afraid of people who are different from us, the others. We’re worried about the economic climate of our city. Maybe we’re just anxious and we don’t know why. The Gospel of John records that the disciples met together on the first Easter Day. They had heard reports that the tomb of Jesus was empty, but they had no proof.
