A binding-to-release strategy for targeted anticancer drug delivery
Abstract Drug conjugates, such as antibody–drug conjugates (ADCs) and small molecule–drug conjugates (SMDCs), are often dependent on efficient receptor-mediated endocytosis for payload release1,2,3—supported by about 10% of targets4,5,6,7. For poorly internalizing targets, drug conjugates dissociate and clear rapidly, limiting efficacy.
Virus-like particles enable targeted gene engineering and pooled CRISPR screening in primary human myeloid cells
|
Abstract Primary human myeloid cells hold promise for immunotherapies, yet efficient, scalable technologies for engineering and screening in these cells remain limited. Here we present a virus-like particle (VLP)-based toolkit that delivers diverse CRISPR editing modalities to human monocytes, macrophages and dendritic cells with high efficiency while preserving viability and innate immune responsiveness.

Nano-enabled spatially selective protein degradation modulates lactate metabolism to potentiate antitumor immunity in liver cancer
Abstract Dysregulated cancer metabolism, driven in part by excessive lactate export through monocarboxylate transporters 1 (MCT1) and 4 (MCT4), generates an acidic tumour microenvironment that suppresses antitumour immunity and diminishes therapeutic efficacy. Although small-molecule inhibitors targeting MCT1 and/or MCT4 have been explored, their clinical translation is limited by systemic toxicity.

Programmable epigenome editing by transient delivery of CRISPR epigenome editor ribonucleoproteins
Abstract Programmable epigenome editors modify gene expression in mammalian cells by altering the local chromatin environment at target loci without inducing DNA breaks. However, the large size of CRISPR-based epigenome editors poses a challenge to their broad use in biomedical research and as future therapies.
Deciphering photocurrent mechanisms at the nanoscale in van der Waals interfaces for enhanced optoelectronic applications
Abstract Integrated photodetectors are vital for their high speed, sensitivity, and low power consumption, with photocurrent driven by the photovoltaic and photothermoelectric (PTE) effects. At the nanoscale, these mechanisms overlap, complicating their separate evaluation.
Aptamer-Enabled Discovery and Clinical Analysis of Exosomal Surface Biomarkers in Hepatocellular Carcinoma
Abstract Exosomal membrane proteins hold promise as cancer biomarkers but face detection challenges due to low abundance and conformational sensitivity. We developed AptEx-ID, a microbead-displayed aptamer platform enabling high-throughput screening of hepatocellular carcinoma (HCC)-specific exosomal surface proteins. Aptamers, selected via competitive binding to tumor-derived versus healthy exosomes, demonstrated high affinity and specificity.

Conformational landscapes of a class I ribonucleotide reductase complex during turnover reveal intrinsic dynamics and asymmetry - Nature Communications
Abstract Understanding the structural dynamics associated with enzymatic catalysis has been a long-standing goal of biochemistry. With the advent of modern cryo-electron microscopy (cryo-EM), it has become conceivable to redefine a protein’s structure as the continuum of all conformations and their distributions. However, capturing and interpreting this information remains challenging.

Trajectory Planning Method for Formation Rendezvous of Underactuated Multi-UUV Under Multiple Constraints
1. Introduction In recent years, with the increasing demands for ocean exploration, resource surveying, and military missions, the application of multiple UUVs in underwater environments has become increasingly widespread [1,2]. Multiple UUVs systems have demonstrated significant advantages in terms of functional redundancy, cooperative operations, and flexibility, gradually becoming a prominent area of current research [3].

CRISPRoff epigenetic editing for programmable gene silencing in human cells without DNA breaks
Abstract The advent of CRISPR-based technologies has enabled the rapid advancement of programmable gene manipulation in cells, tissues, and whole organisms. An emerging platform for targeted gene perturbation is epigenetic editing, the direct editing of chemical modifications on DNA and histones that ultimately results in repression or activation of the targeted gene.

Conformational Landscapes of a Class I Ribonucleotide Reductase Complex during Turnover Reveal Intrinsic Dynamics and Asymmetry
Abstract Understanding the structural dynamics associated with enzymatic catalysis has been a long-standing goal of structural biology. A wide range of motions, from small side-chain fluctuations to large domain rearrangements, have been implicated in enzyme function by experimental and computational studies. However, because structural techniques generally depend on averaging, direct visualization of conformational landscapes during turnover has been challenging.

Miffi: Improved accuracy of CNN-based cryo-EM micrograph filtering with fine-tuning and Fourier space information
Abstract Efficient and high-accuracy filtering of cryo-electron microscopy (cryo-EM) micrographs is an emerging challenge with the growing speed of data collection and sizes of datasets. Convolutional neural networks (CNNs) are machine learning models that have been proven successful in many computer vision tasks, and have been previously applied to cryo-EM micrograph filtering.

Hierarchical replay of multi-regional sequential spiking associated with working memory
Abstract How does millisecond-scale neural activity mediate behaviors over seconds? We recorded brain-wide activity in mice performing an olfactory working-memory task to decipher cross-region organization of activity. Spike-correlograms revealed millisecond within- and cross-region spike couplings, more prominent among neurons encoding similar memories. Spike coupling linked neurons into motifs of chains, single loops, and nested loops, especially among hippocampal and prefrontal-cortex neurons.
Regression Analysis of Dependent Current Status Data with Left Truncation
1. Introduction Current status data generally arise in demographic, tumorigenic, and epidemiological fields [1,2,3]. One significant feature of the current status data is that the failure time cannot be accurately observed. On the contrary, it is known that the failure time is less than or greater than the observation or examination time. One common feature of studies that produce such data is that participants are only observed once, perhaps due to the limitation of resources.

FusionPillars: A 3D Object Detection Network with Cross-Fusion and Self-Fusion
Abstract : In the field of unmanned systems, cameras and LiDAR are important sensors that provide complementary information. However, the question of how to effectively fuse data from two different modalities has always been a great challenge. In this paper, inspired by the idea of deep fusion, we propose a one-stage end-to-end network named FusionPillars to fuse multisensor data (namely LiDAR point cloud and camera images).
Quantum Control of a Single Magnon in a Macroscopic Spin System
Abstract Nonclassical quantum states are the pivotal features of a quantum system that differs from its classical counterpart. However, the generation and coherent control of quantum states in a macroscopic spin system remain an outstanding challenge. Here we experimentally demonstrate the quantum control of a single magnon in a macroscopic spin system (i.e., 1 mm-diameter yttrium-iron-garnet sphere) coupled to a superconducting qubit via a microwave cavity.

Effects of Layer Thickness on the Residual Stresses of CIGS Solar Cells with Polyimide Substrate
1. Introduction Copper indium gallium diselenide (CIGS) solar cells are one of the most widely used flexible solar cells in the market. They are typically composed of Zinc Oxide as a front contact (ZnO), Cadmium Sulfide as a buffer layer (CdS), CIGS as an absorber layer (CIGS), and Molybdenum as a back contact (Mo). Soda-lime glass (SLG) is a material commonly used as a rigid substrate while stainless steel (SS) and polyimide (PI) are materials commonly used as flexible substrates.
