A transferable gut microbiota–bile acid pathway programs nanomedicine pharmacokinetics and therapeutic response
Abstract The clinical efficacy of nanomedicines is often limited by hepatic sequestration, yet the endogenous programs determining this clearance state remain incompletely understood. Here we identify the gut microbiota as a regulator of nanomedicine biodistribution through bile-acid-associated programming of Kupffer cell phagocytic state.
Microdosimetric characterization of proton relative biological effectiveness in pancreatic ductal adenocarcinoma cell lines
Abstract Purpose: Pancreatic ductal adenocarcinoma (PDAC) is among the most radioresistant malignancies. Proton therapy assumes a uniform relative biological effectiveness (RBE) of 1.1 regardless of radiation quality. We characterized proton RBE as a function of dose-mean lineal energy (ȳD) in PDAC cell lines. Methods: Proton RBE was assessed in Panc-1 and COLO 357 FG cells using a high-throughput 96-well irradiation platform with an 81.4 MeV spot-scanning proton beam.
Spatial multi-omics landscape of colorectal cancer macro- and micrometastases

Introduction While early detection through screening has significantly reduced the incidence of localized colorectal cancer (CRC),1 limited understanding of the biological mechanisms driving colorectal metastasis remains a major barrier to improving long-term survival.

Acquired resistance in cancer: towards targeted therapeutic strategies - Nature Reviews Cancer
|
Abstract Development of acquired therapeutic resistance limits the efficacy of cancer treatments and accounts for therapeutic failure in most patients. How resistance arises, varies across cancer types and differs depending on therapeutic modalities is incompletely understood. Novel strategies that address and overcome the various and complex resistance mechanisms necessitate a deep understanding of the underlying dynamics.

Radiotherapy promotes cuproptosis and synergizes with cuproptosis inducers to overcome tumor radioresistance
|
Results The involvement of cuproptosis in RT remains unclear. We observed a dose-dependent increase in intracellular copper levels following RT in cancer cells (Figure 1A), suggesting a potential link between RT and cuproptosis. The inductively coupled plasma mass spectrometry (ICP-MS) analysis further confirmed that RT induced an increase in copper concentrations in cancer cells (Figure 1B).

"IRE1α Determines Ferroptosis Sensitivity Through Regulation of Glutath" by Dadi Jiang, Youming Guo et al.
Abstract Cellular sensitivity to ferroptosis is primarily regulated by mechanisms mediating lipid hydroperoxide detoxification. We show that inositol-requiring enzyme 1 (IRE1α), an endoplasmic reticulum (ER) resident protein critical for the unfolded protein response (UPR), also determines cellular sensitivity to ferroptosis. Cancer and normal cells depleted of IRE1α gain resistance to ferroptosis, while enhanced IRE1α expression promotes sensitivity to ferroptosis.

Synthetic cationic helical polypeptides for the stimulation of antitumour innate immune pathways in antigen-presenting cells - Nature Biomedical Engineering
|
Abstract Intracellular DNA sensors regulate innate immunity and can provide a bridge to adaptive immunogenicity. However, the activation of the sensors in antigen-presenting cells (APCs) by natural agonists such as double-stranded DNAs or cyclic nucleotides is impeded by poor intracellular delivery, serum stability, enzymatic degradation and rapid systemic clearance.
